  ThematrixoftheLSColumnsiscomposedofferromagneticspheres,whicharecoveredwithacell-friendlycoatingallowingfastandgentleseparationofcells.WhenplacedinthemagneticfieldofaMACS^?Separator,thespheresamplifythemagneticfieldby10,000-fold,thusinducingahighgrADIentwithinthecolumn.ThisiscrucialforisolationofcellswhichareonlyminimallylabeledwithMACS^?MicroBeads,leavingenoughepitopesfreeforconcurrentantibodystaining.Thespacebetweenthespheresisseveraltimeslargerthanprimaryandmostculturedcells.Thisallowsthecellstofreelyflowthroughthecolumn.MagneticallylabeledcellsareheldinsUSPensionwithinthecolumnanddonotactually“bind”thecolumnmatrix.Thissuspensionminimizesstressonthecellsandallowsforefficientsterilewashingbyavoidingcellaggregation.
