TSKgelChelate-5PWcolumnsutilizethetendencyofiminodiaceticacid(IDA)toformacomplexwithmetalions,suchasZn2+,Ni2+andCu2+.Thecolumnisloadedwithdivalentmetalionsbychelation.PeptidesandproteinscontaininghistidineresidueswillnormallyadsorbtothesechelatedionsatneutralpH.Theretainedcompoundscanbeelutedwithbuffercontainingimidazoleorglycine.Thekeytomakingsuccessfuluseofthisretentionmechanismistheproperselectionofmetalionsforchelationandtheelutionbuffertodesorbtheanalytes.Ingeneral,Cu2+,interactsbetterwithproteins;however,resolutionisusuallyenhancedwithZn2+.AgrADIentmobilephasecontainingincreasingimidazoleorglycineconcentrationsisusedtoelutetheretainedcompounds.Alternatively,adecreasingpHgradientcanbeusedforelutionpurposesaswell.
